Background Principal Names: Progesterone receptor Official Gene Symbol: PGR Gene ID: 5241 (Human) Gene Map Locus: 11q22-q23 (Human) The human progesterone receptor (hPR) belongs to the superfamily of nuclear receptors (including those for steroid and thyroid hormones, and retinoids) whose members coordinate morphogenesis and homeostasis in response to the binding of their ligands. These receptors act as transcriptional factors that regulate gene expression positively or negatively by interacting with cognate DNA sequences. It exists as two functionally distinct isoforms, PR-A and PR-B. The proteins are identical except for an additional 164 residues located at the N terminus of PR-B. whereas both isoforms appear to have similar DNA binding affinities, PR-B in fact has a greatly increased intrinsic binding affinity and cooperative binding ability relative to PR-A. The differences map to a unique AF3 in the B-upstream segment (BUS), at the far N terminus of PR-B, which is missing in PR-A. It maps to 11q22-q23 region of human chromosome. |

Application Notes We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Specificity: This antibody is specific to human progesterone receptor protein. Progesterone receptor is an estrogen regulated protein and its expression indicates a responsive estrogen receptor pathway. Cellular Localization: Nuclear. |
