| Background Principal Names: Tumor protein p53 (Li-Fraumeni syndrome) Official Gene Symbol: TP53 Gene ID: 7157 (Human) Gene Map Locus: 17p13.1 (Human) Tumor protein p53, a nuclear protein, plays an essential role in the regulation of cell cycle, specifically in the transition from G0 to G1.It is a DNA-binding protein containing DNA-binding, oligomerization and transcription activation domains. It is postulated to bind as a tetramer to a p53-binding site and activate expression of downstream genes that inhibit growth and/or invasion, and thus function as a tumor suppressor. The gene is 20 kilobases (kb) long and has 11 exons, the first and second exons being separated by an intron of 10 kb. Mutations in the evolutionarily conserved codons of the p53 tumor suppressor gene are common in diverse types of human cancer which includes cancer of colon, lung, esophagus, breast, liver, brain, reticuloendothelial tissues, and hemopoietic tissues. It maps to the 17p13.1 region of human chromosome. |

| Application Notes We suggest an incubation period of 30 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user. Formalin fixed paraffin embedded tissue sections require high temperature antigen unmasking with 10 mM citrate buffer, pH 6.0 prior to immunostaining. Specificity: This antibody is specific to human p53 protein, both wild and mutant forms under denaturing and non-denaturing conditions. It maps the n-terminus region of the p53 molecule in the region of amino acids 20 and 25. Cellular Localization: Nuclear. |
