-20°CGrowth factor active in angiogenesis, vasculogenesis and endothelial cell growth. Induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. NRP1/Neuropilin-1 binds isoforms VEGF-165 and VEGF-145. Isoform VEGF165B binds to KDR but does not activate downstream signaling pathways, does not activate angiogenesis and inhibits tumor growth.
Applications:
Suitable for use in ELISA, Western Blot and Neutralization. Other applications not tested.
Recommended Dilution:
ELISA: In a sandwich ELISA (assuming 100ul/well), a concentration of 2-4ug/ml of this antibody will detect at least 100pg/ml of recombinant human VEGF165 when used with biotinylated antigen affinity purified anti-human VEGF165 as the detection antibody at a concentration of approximately 0.25-1ug/ml.
Western Blot: To detect hVEGF165 by Western Blot analysis this antibody can be used at a concentration of 0.25-0.5ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hVEGF165 is 0.25-0.5ng/lane, under non-reducing conditions.
Neutralization: To yield one-half maximal inhibition of the biological activity of Human VEGF (10ng/ml), a concentration of 0.2ug/ml of this antibody is required.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
