Alkaline phosphatase (AP) conjugated anti-horse primary antibodies are enzyme-labeled immunoreagents for colorimetric, chemiluminescent, and fluorescent detection of equine antigens in applications requiring prolonged signal stability. Produced by immunizing hosts such as goat or rabbit with purified horse immunoglobulins, the antibodies are affinity-purified and conjugated to alkaline phosphatase, a 140 kDa metalloenzyme derived from calf intestine or bovine tissues. AP catalyzes the hydrolysis of phosphate esters, enabling detection with substrates such as BCIP/NBT (producing a purple-blue precipitate), pNPP (yellow soluble product), or CDP-Star® (chemiluminescent emission). Unlike HRP, AP offers the advantage of continuous signal generation without substrate depletion and exhibits greater stability in the presence of azides and other preservatives.
Applications
AP conjugates are widely employed in ELISA, where pNPP provides sensitive colorimetric quantification of equine antibody titers. In immunohistochemistry and in situ hybridization, BCIP/NBT precipitates yield permanent chromogenic signals suitable for long-term slide archival. Western blotting applications benefit from chemiluminescent AP substrates such as CDP-Star® or APS-5, enabling detection of low-abundance equine proteins. Additional applications include dot blots, tissue microarrays, and membrane-based assays where background interference from endogenous peroxidases is a concern.
Performance and Recommended Use
Typical working dilutions range from 1:500 to 1:10,000 depending on the substrate, assay format, and experimental conditions. Alkaline phosphatase conjugates are particularly advantageous for applications requiring sustained signal development and high sensitivity. These reagents are intended for research use only (RUO) and are not designed for use in diagnostic procedures.
