Calcium hydride

Calcium hydride serves as a powerful desiccant and reducing agent in anhydrous biochemical protocols. It is particularly used for solvent drying prior to moisture-sensitive reactions such as Grignard formations and hydride-mediated reductions in nucleotide synthesis, where strict control of water content is essential for reaction efficiency and reproducibility.

Chemical Properties

Calcium hydride (CaH₂, MW 42.09 g/mol) is a gray orthorhombic powder with a density of 1.70 g/cm³ and a melting point of 816°C, decomposing above 1000°C. It exhibits a saline hydride structure analogous to the cotunnite-type lattice, in which Ca²⁺ cations are coordinated by eight H⁻ anions in a distorted cubic geometry (Ca–H distance ≈ 2.42 Å). Insoluble in most organic solvents, it reacts vigorously with water according to the reaction CaH₂ + 2H₂O → Ca(OH)₂ + 2H₂, releasing hydrogen gas with an enthalpy change of ΔH = −177 kJ/mol. It remains stable under inert atmospheres up to approximately 400°C.

Applications in Biochemical and Organic Synthesis

In organic synthesis, calcium hydride is commonly used at concentrations of 1–5 wt% for drying solvents such as tetrahydrofuran (THF), diethyl ether (Et₂O), and dioxane, typically under nitrogen atmosphere with stirring for 24–48 hours. This process efficiently removes residual water, alcohols, and amines to levels below 10 ppm, which is critical for moisture-sensitive reactions including organolithium additions to carbonyl compounds and silyl protection strategies.

In peptide chemistry, calcium hydride is applied for the pre-treatment of solvents such as dimethylformamide (DMF) used in Fmoc-based coupling reactions, helping to prevent side reactions such as aspartimide formation. In molecular biology workflows, it is also employed to purify solvents used in phosphoramidite oligonucleotide synthesis, where maintaining water content below 5 ppm is essential to achieve coupling efficiencies exceeding 98%.
