For the quantitative determination of rat NAD-dependent protein deacetylase sirtuin-1 (SIRT1) in serum, plasma, tissue homogenates, and cell lysates.

Specificity:
This assay has high sensitivity and excellent specificity for detection of rat SIRT1. No significant cross-reactivity or interference between rat SIRT1 and analogues was observed.

Assay Principle:
This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SIRT1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SIRT1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SIRT1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SIRT1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Storage and Stability:
Store all kit components at 4°C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
