Keratan sulfate (also known as keratosulfate) is a widely distributed glycosaminoglycan in tensional and weight-bearing connective tissues, epithelial tissues, and the central and peripheral nervous system. It is attached to a number of proteoglycan and glycoprotein core proteins. Three types of keratan sulfate have been described: KS I, KS II, and KS III depending on their distribution pattern. Keratan sulfate is the only glycosaminoglycan (GAG) that does not contain uronic acid; therefore, it is not cleavable by the eliminative enzymatic cleavage. The biosynthesis of keratan sulfate occurs in two stages. In the first stage, the region binding the core protein with GAG is created followed by the elongation of the chain and its modification. The elongation of keratan sulfate chains of all types occurs by alternate attaching of Gal and GlcNAc residues, catalyzed by β-1,4-galactotransferase and β-1,3-N-acetyloglucosaminotransferase, respectively. Sulfation of keratan sulfate is carried out by at least two sulfotransferases. Gal 6-O-sulfotransferase transfers sulfate to position 6 of Gal residues located in the internal region of keratan sulfate and GlcNAc 6-O-sulfotransferase transfers sulfate to position 6 of GalNAc residues located at the non-reducing terminal of keratan sulfate.

Applications:
Suitable for use in Western Blot, ELISA, Immunohistochemistry and Immunoprecipitation. Other applications not tested.

Recommended Dilutions:
Immunohistochemistry (paraffin): 1:50 detected Keratan Sulfate in human cartilage tissue sections using heat-induced epitope retrieval (HIER).
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
