The cytotoxicity test with crystal violet (CV) is one of the commonly used methods to detect cell viability or cytotoxicity of drugs.
Crystal violet is a triarylmethane dye that can bind to ribose-like molecules such as DNA in nuclei. Normally, dead adherent cells will detach from the cell culture plates and be removed from the viable cell population during the washing steps. CV staining can be used to quantify the total DNA of the remaining population and thereby determine cell viability. CV staining is directly proportional to cell biomass and can be measured at 570 nm. CV staining is a rapid and versatile test for screening for cell viability under various stimulation or inhibition conditions. The cytotoxicity test kits for CV cells also include doxorubicin as a positive control. This type of kit offers an excellent and effective method for in vitro cytotoxicity studies as well as for screening of high-throughput drugs.
