Assay Principles

  • Colorimetric Assays: Glucose oxidase catalyzes the oxidation of β-D-glucose to gluconic acid and H2O2. In the presence of peroxidase (POD), H2O2 reacts with chromogenic substrates (e.g., 4-aminoantipyrine and phenol) to form a colored quinoneimine dye measurable at 505–570 nm. Mutarotase is often included to convert α-D-glucose to β-D-glucose, ensuring full glucose detection.
  • Fluorometric Assays: The generated H2O2 reacts with a fluorogenic probe in the presence of peroxidase, producing fluorescence measurable at excitation/emission wavelengths around 535/587 nm, offering higher sensitivity.
