Applications ELISA, ID, IF, IHC, IP
Recommended Dilution Can be used in precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgD in mouse serum and other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of mouse IgD from serum or plasma. This antiserum is not intended for use in non-precipitating antibodybinding or other highly sensitive assays. For such use labelled an unlabelled cytochemical and immunoassay grade reagents meeting the necessary additional specificity an performance requirements are available.
Antibody titre: Precipitin titre 1/32 when tested against monoclonal IgD containing mouse serum in agarblock immunodiffusion titration.
Reactivities Mouse
Host Goat
Clonality Polyclonal
Immunogen Highly purified polyclonal and monoclonal IgD isolated from Balb/C and C57Bl serum.
freund’s complete adjuvant is used in the first step of the immunization procedure.
Specificity The reactivity of the antiserum is restricted to the Fc portion of the IgD molecule, which represents strict isotype (class specificity. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against mouse serum belonging to different allotypic groups, a single characteristic precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with the purified IgD from Balb/C and C57Bl. No precipitation reaction is obtained with purified IgG, including all subclasses, IgA, IgM, IgG/Fab fragments or other serum proteins. The antiserum also reacts with membranebound IgD in peripheral blood cells of different mouse strains as tested by immunofluorescence microscopy.
Cross-reactivity: The antiserum does not cross-react with any other component of the mouse Ig system. Inter-species cross reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants, but this has not been tested because of the very low level of IgD in most species.
Formulation State: Serum
State: Lyophilized (Delipidated and heat inactivated) stable whole antiserum - No preservative added
Form Serum
Reconstitution Method Restore by adding 1.0 ml sterile distilled water.
Dilutions may be prepared by adding PBS, pH 7.2
Concentration Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.
Conjugation Unconjugated
Storage Store lyophilized at 2-8°C for 6 months or at -20°C long term.
After reconstitution store the antibody undiluted at 2-8°C for one month
or (in aliquots) at -20°C long term.
Avoid repeated freezing and thawing.
Note Adsorption Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other components of the immunoglobulin system or reacting with other serum proteins. Special attention is given to the elimination of antibodies to he common Ig/Fab portion. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
