Aristolochic acid III is a naturally occurring nitrophenanthrene carboxylic acid and a structural isomer of aristolochic acid I, frequently found as a minor contaminant in Aristolochia herbal extracts. It acts as a potent DNA-damaging agent, forming both 2′-deoxyguanosine and 2′-deoxyadenosine adducts, with dose-adjusted adduct levels 30 to 80 times higher than those of aristolochic acid I in the kidney and urinary bladder. This compound exhibits strong organotropism for the urinary system, where its DNA adduct formation is uniquely elevated compared to other related genotoxicants.
