| In vitro | METHODS: Three different tumor cell lines, HeLa, MDA-MB231 and Jurkat T cells, were incubated with Cycloheximide (2.5 µM) for 24 hours. The inhibitory effects of the compounds on translation and transcription were detected by metabolic labeling. RESULTS: The IC50 values of Cycloheximide for protein synthesis and RNA synthesis in vivo were 532.5 nM and 2880 nM, respectively. [1] |
| In vivo | METHODS: To investigate the effect of Cycloheximide on the latency of the memory test in mice, 30, 60 or 120 mg/kg of Cycloheximide was injected before training with 200 μA electric shock. RESULTS: Cycloheximide has a significant effect on the latency of the memory test. In the control group, the delay time of the test was significantly higher than that of the training. The test of the lowest dose of Cycloheximide, namely 30 mg/kg, resulted in a significantly higher latency of the test than that of the control group. After receiving two higher doses of Cycloheximide, the performance of either mouse in the experiment was comparable to that of the control group, and the higher doses did not impair memory under these conditions, thus forming an inverted U-shaped dose-response curve. For example, if the intended dosage is 10 mg/kg for animals weighing 20 g , with a dosing volume of 100 μL per animal, and a total of 10 animals are to be administered, using a formulation of 
10% DMSO+ 40% PEG300+ 5% Tween 80+ 45% Saline/PBS/ddH2O , the resulting working solution concentration would be 2 mg/mL.Dissolve 2 mg of the compound in 100 μL DMSO to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
