Pertuzumab is a humanized monoclonal antibody targeting HER2. It binds to the extracellular dimerization domain of the HER2 protein, preventing HER2 from forming heterodimers with other HER family receptors and thereby blocking tumor cell growth and survival signaling pathways. Pertuzumab can be used in research on HER2-positive breast cancer.

Method: MDA-175 breast cancer cells were treated with pertuzumab at concentrations ranging from 0 to 100 μg/mL for 24 h.
Result: Pertuzumab synergistically inhibited cell proliferation and enhanced caspase-3/7 activity as early as 24 h after treatment. ^[1]
Method: SK-BR-3 and KPL-4 breast cancer cells were treated with pertuzumab at a concentration of 15μg/mL before stimulation with 1 nmol/L NRG-1b for 15minutes or 24 h, followed by western blot assay.
Result: Pertuzumab inhibited NRG-1β-stimulated HER3 and AKT phosphorylation in SK-BR-3 cells and this inhibitory effect was more pronounced after prolonged incubation (24 h).^[1]

Method: Mice bearing MDA-175 tumors received a single intraperitoneal injection of pertuzumab (3 mg/kg), T-DM1 (10 mg/kg), or the combination of both agents.
Result: The combination of pertuzumab and T-DM1 significantly prolonged the time to tumor doubling compared with either single-agent treatment or vehicle control groups (P < 0.0001 for combination vs. vehicle or pertuzumab; P = 0.0003 for combination vs. T-DM1). ^[1]
