Caproic acid

Caproic acid is a medium-chain fatty acid (C6:0) produced through gut microbial fermentation. It exhibits significant antimicrobial activity and is rapidly metabolized into ketone bodies, contributing to energy homeostasis, particularly under fasting conditions.

Chemical Properties

Caproic acid (CH₃(CH₂)₄COOH, molecular weight 116.16 g/mol), also known as hexanoic acid, appears as a colorless to pale yellow oily liquid characterized by a rancid, goat-like odor. It has a density of 0.925 g/cm³, a boiling point of 205°C, and a melting point of -3°C. As a moderately acidic compound (pKa ≈ 4.85), it exhibits limited solubility in water (10.9 g/L) but is readily soluble in organic solvents such as ethanol and ether. Its volatility under steam conditions makes it suitable for GC-MS analysis, and it presents a flash point of 102°C. Metabolically, caproic acid undergoes β-oxidation to yield three acetyl-CoA molecules, corresponding to an energy production of approximately 17 ATP, and can also undergo esterification to form flavor compounds.

Biochemical Applications

In microbiome research, caproic acid is typically used at concentrations ranging from 0.5 to 2 mM to disrupt pathogenic biofilms, including those formed by Clostridium difficile and Candida albicans, primarily through membrane permeabilization mechanisms while preserving commensal microbial populations. In metabolic studies, medium-chain fatty acids such as caproic acid are preferentially oxidized in liver mitochondria, where they are converted into ketone bodies, providing an alternative energy source for neuronal tissues, particularly in conditions such as hypoalbuminemia. Additionally, in protein engineering, caproic anhydride is utilized for Nε-acylation of lysine residues, enabling modulation of protein stability and aggregation properties. Antimicrobial activity is quantitatively assessed through minimum inhibitory concentration (MIC) assays, typically ranging from 0.5 to 2 mM against Gram-positive bacteria and fungal species.
