• Signal-to-noise ratio with mammalian cells.

• Consistency of fluorescence and absorbance readouts.

• Absence of contamination.

• Optimize cell density and incubation time for each cell type to ensure that the signal remains within the linear detection range.
• Include untreated cell controls, medium-only blanks and compound-only controls when testing coloured, fluorescent or reducing substances.
• Compounds with intrinsic absorbance, fluorescence or reducing activity may interfere with resazurin conversion and should be evaluated using appropriate controls.
• Avoid prolonged incubation, as complete reduction of resazurin may reduce assay linearity and compromise quantitative interpretation.
• Protect the reagent from prolonged exposure to light and use the same incubation conditions across all samples and controls.
