ELISA Kit Principle | This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Ethinylestradiol (EE) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Ethinylestradiol (EE) and unlabeled Ethinylestradiol (EE) (Standards or samples) with the pre-coated antibody specific to Ethinylestradiol (EE). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Ethinylestradiol (EE) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Ethinylestradiol (EE) in the sample. | ||||||||||||||||||||||||

ELISA Kit Reproducibility | Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Ethinylestradiol (EE) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Ethinylestradiol (EE) were tested on 3 different plates, 8 replicates in each plate. Description of Target | Ethinyl Estradiol is a semisynthetic estrogen. Ethinyl estradiol binds to the estrogen receptor complex and enters the nucleus, activating DNA transcription of genes involved in estrogenic cellular responses. This agent also inhibits 5-alpha reductase in epididymal tissue, which lowers testosterone levels and may delay progression of prostatic cancer. In addition to its antineoplastic effects, ethinyl estradiol protects against osteoporosis. In animal models, short-term therapy with this agent has been shown to provide long-term protection against breast cancer, mimicking the antitumor effects of pregnancy. (NCI04) |
