Description

PAMP-12 (unmodified) is an endogenous peptide and is a MrgX2 agonist. PAMP-12 (unmodified) can reduce cAMP accumulation, increase Ca2+ levels, enhance beta-arrestin recruitment, decrease IP-1, and increases phosphoERK. PAMP-12 (unmodified) can elicit hypotension through inhibiting catecholamine secretion from sympathetic nerve endings and adrenal chromaffin cells. PAMP-12 (unmodified) can be used for the research of hypotension and ulcerative colitis[1][2].

In Vitro

PAMP-12 (10-9-10-6 M; 30 min) potently inhibits Forskolin (HY-15371)-elevated cAMP accumulation in CHO-MrgX2 cells with an EC50 of 57.2 nM[1].
PAMP-12 (10-10-10-6 M) potently induces intracellular Ca2+ mobilization in CHO-MrgX2 cells with an EC50 of 41.0 nM[1].
PAMP-12 (10-10-10-6 M; 60 min) directly binds to and activates MrgX2 in HEK293 cell membranes, stimulating [35S]GTPγS binding with an EC50 of 20.8 nM[1].
PAMP-12 (90 min) stimulation induces greater β-arrestin recruitment to the MRGPRX2 62Ser variant[2].
PAMP-12 (10-7-10-4 M; 2 h) stimulation results in greater IP-1 accumulation, indicating stronger Gq/phospholipase C pathway activation, in CHO cells expressing the MRGPRX2 Asn62 variant compared to the 62Ser variant[2].
PAMP-12 (10-20 μM; 0.5-5 min) stimulation induces greater pERK activation in HMC-1.1 cells expressing the MRGPRX262Ser variant compared to the Asn62 variant[2].

Cell Line: HMC-1.1 human mast cells stably expressing MRGPRX2 Asn62 or 62Ser variants
Concentration: 10 μM; 20 μM
Incubation Time: 0.5, 1, 5 minutes
Result: Reached maximal pERK activation at 0.5 and 1 minutes for both variants.
Showed significantly higher normalized pERK levels in the 62Ser variant compared to the Asn62 variant at 1 minute post-stimulation for both 10 μM and 20 μM concentrations.
