Affinity purified Lotus tetragonolobus Lectin (LTL) is immobilized to Agarose following CNBr coupling. The final product is stabilized in a buffer to retain maximum activity.

Carbohydrate Specificity:
alpha-Fucose

Conjugate Specificity:
Agarose Beads

Inhibitory Carbohydrate:
50-100mM L-Fucose

Lectin Activity:
Less than 10ug/ml will agglutinate type O human erythrocytes.

Protocol:
All materials should be equilibrated to the temperature at which the chromatography will be performed.

Binding:
1. Equilibrate the column with 0.1M HEPES buffer containing 0.15M sodium chloride, 0.1mM CaCl2, 0.1mM MnCl2, pH 7.5.

2. Dilute the sample 1:10 in same equilibration buffer (100ul in 900ul) and load to the column allowing to circulate for 1 hour or incubate in a tube in a rotating motion for an hour at RT.

3. Wash unbound material from the column with 4 volumes of equilibration buffer/resin.

Elution:
1. Elute the bound glycoprotein in the same equilibration buffer with the addition of 50-200mM L-Fucose.

Column Regeneration and Storage:
1. Wash the gel with about 4-6 column volumes of 1.5M sodium chloride and then distilled water.

2. Equilibrate the column with 0.5M sodium chloride and 0.02% sodium azide prior to storage.

Storage and Stability:
May be stored at 4°C. For long-term storage, aliquot and store at 4°C. Do not freeze. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
