• Modeling Mechanism：

  Dextran (MW 5000) induces mast cell degranulation and release of inflammatory mediators such as histamine and serotonin, increasing local vascular permeability and causing plasma components to leak into the interstitial tissue, thus triggering acute edema of the foot tissue.

• Modeling Method：

  Experimental Subject：

  Albino rats (170–200 g)

  Dosage and Administration Route：

  0.1 mL of freshly prepared 1% dextran solution, administered via intra‑plantar injection into the right hind paw.

  Dosing Frequency and Duration Model：

  Single dose

• Validation：

  After administration, foot swelling gradually worsened, exhibiting characteristics of acute inflammatory edema, and the increase in foot volume was significantly higher than that in the control group (P<0.05).

*Precautions： On the final day of the experiment, 0.1 mL of a freshly prepared 1% dextran solution was injected into the plantar surface of each rat’s right hind paw. The plantar volume was measured before administration and at specified time intervals (every 30 minutes over a 4-hour period) after administration. Changes in plantar volume were determined using a vernier caliper, and anti-inflammatory activity was calculated.
