Annexin V Conjugates
Annexin V fluorescent conjugates can be used to detect apoptotic cells. Annexin V is available conjugated to a wide variety of dyes.

Flow cytometry analysis of untreated and staurosporine-treated Jurkat cell stained with NucView® 488 Caspase-3 Substrate and CF®647 Annexin V.
Jurkat cells were treated with staurosporine to induce apoptosis, or with DMSO as a negative control for the times indicated, then stained for 15 minutes at room temperature with NucView® 530 Caspase-3 Substrate and CF®640R Annexin V in cell culture medium prior to flow cytometry analysis on a BD LSRII flow cytometer.
Jurkat cells were treated with DMSO (negative control, blue) or 0.1 uM, 0.25 uM, or 1 uM staurosporine (red) to induce apoptosis in the presence of NucView® 530 and CF®405M Annexin V for four hours at 37°C before analysis on a BD LSR II flow cytometer. Staining with NucView® 530 and CF®405M Annexin V increase with increasing concentration of staurosporine.
Jurkat cells were treated with 1 uM staurosporine for 4 hours, then treated (green histogram) and untreated (gray histogram) cells were each stained with 2.5 ug/mL CF®488A Annexin V and analyzed by flow cytometry in the B515 (FITC) channel.
Jurkat cells were treated with 1 uM staurosporine for 4 hours, then treated (pink histogram) and untreated (gray histogram) cells were each stained with 5 uL APC Annexin V and analyzed by flow cytometry in the R660 (APC) channel.
Jurkat cells were treated with 1 uM staurosporine for 4 hours, then treated (orange histograms) and untreated (gray histograms) Jurkats were each stained with 5 uL R-PE Annexin V and analyzed by flow cytometry in the Y585 (PE) channel.
Jurkat cells were treated with 1 uM staurosporine for 4 hours, then treated (teal filled histogram) and untreated (gray histogram) Jurkats were each stained with 2.5 ug/mL CF®740 Annexin V and analyzed by flow cytometry in the R637 (APC-750) channel. Unstained cells, both treated and untreated, are shown in black.

Annexin V fluorescent conjugates can be used to detect apoptotic cells. The human anticoagulant Annexin V is a 35-36 kDa calcium-dependent phospholipid-binding protein with high affinity for phosphatidylserine (PS). In normal viable cells, PS is located on the inner leaflet of the cytoplasmic membrane. In apoptotic cells, however, PS is translocated from the inner to the outer leaflet of the plasma membrane, where it can be detected by fluorescently labeled Annexin V.

CF® Dye, biotin, and FITC conjugates of Annexin V are provided as 50 ug/mL solutions in 10 mM Tris, 1 mM EDTA, 30 mM NaCl, 1 mg/mL bovine serum albumin, 0.1% sodium azide, pH 7.5. R-PE and APC Annexin V conjugates are supplied in PBS, 5 mg/mL BSA, 0.05% sodium azide at 5 uL per test.

Note: Conjugates of blue-fluorescent dyes like CF®350, CF®405S and CF®405M are not recommended for detecting low abundance targets and may be challenging to use in tissue specimens. Blue dyes have lower fluorescence and photostability, and cells and tissue have high autofluorescence in blue wavelengths, resulting in lower signal to noise compared to other colors.
