Human CD81 is a member of the tetraspan family of leukocyte cell surface molecules which are involved in signal transduction, cell-cell adhesion, and cellular activation (1). Association of CD81 with integrins appears to be functionally important in the regulation of cell adhesion and activation (2, 3). Recent work with CD81-null mice indicates CD81 is important for development of humoral immune response (4).

Applications:
Suitable for use in Flow Cytometry and Western Blot. Other applications not tested.

Recommended Dilutions:
Flow Cytometry: 1:50 (10ug/ml). 80ul labels 5x10e5 cultured human CEM cells.
Optimal dilutions to be determined by the researcher.

Flow Cytometry Protocol:
1. 5x10e5 cultured human CEM cells were washed and incubated 45 minutes on ice with 80ul of C2433-10C at a 1:50 dilution factor (10ug/ml).
2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 2.1 log10 fluorescent units when compared to a Mouse IgG1/FITC negative control at a similar concentration.
4. Binding was blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml anti-CD81 antibody (C2433-10A).

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. FITC conjugates are sensitive to light.
