Product Categories/Family for anti-HIST4H4 antibody

Antibodies; Abs to Histones
Applications Tested/Suitable for anti-HIST4H4 antibody
Western Blot (WB), Immunoprecipitation (IP)
Application Notes for anti-HIST4H4 antibody
Western Blotting: 1:2000 dilution detects acetylated histone H4 in Tetrahymena macronuclei and acid extracted proteins from HeLa cells treated with 5mM sodium butyrate. Sodium butyrate, an inhibitor of deacetylases, was used to enhance detection of acetylated histone H4., Chromatin Immunoprecipitation: 5-10ul immunoprecipitates transcriptionally active, chromatin containing acetylated histone H4 from 2X 106 serum stimulated HeLa cells., Immunocytochemistry: Not recommended.

Western Blot (WB) of anti-HIST4H4 antibody

WB: Acid extract of untreated (lane 1) or butyrate-treated (lane 2) HeLa cells probed with MBS616728 Histone H4 at 1:2000 dilution.
Rabbit Anti-HIST4H4 Polyclonal Antibody – Cat# MBS616728

Immunoprecipitation (IP) of anti-HIST4H4 antibody

Sonicated chromatin prepared from untreated HeLa cells (1x10e6 cell equivalents) was subjected to chromatin immunoprecipitation using 4uL of either a normal rabbit antiserum or Acetyl Histone H4 serum and the Magna ChIP A Kit. Successful immunoprecipitation of acetyl histone H4 associated DNA fragments was verified by qPCR using control ChIP Primers flanking the human GAPDH promoter.

Other Notes

Small volumes of anti-HIST4H4 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for HIST4H4

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. [provided by RefSeq]

UniProt Comments for HIST4H4

H4: histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The gene encoding H4 is intronless.
