Human whole blood collected in Acid-Citrate-Dextrose Solution A (ACD-A) is a minimally processed peripheral blood specimen containing erythrocytes, leukocytes, platelets, plasma proteins, metabolites, and other circulating components. ACD-A contains citric acid, sodium citrate, and dextrose. Citrate provides anticoagulation primarily by chelating ionized calcium required for coagulation, while dextrose serves as a metabolic substrate.
For research, ACD-A–anticoagulated whole blood provides access to circulating cellular and soluble components without prior separation into blood fractions. Its use has been described in whole-blood gene-expression and transcriptomic studies, PBMC isolation, leukocyte-based assays, immunological investigations, hematology, and molecular research. Because anticoagulants and pre-analytical conditions can influence cellular function and molecular measurements, their compatibility should be considered for the intended downstream assay.
