Kinyoun stain

Kinyoun staining is a cold acid-fast staining (AFB) technique widely used in clinical microbiology, pathology, and research laboratories for the detection of acid-fast microorganisms, particularly Mycobacterium species and partially acid-fast bacteria such as Nocardia. Unlike the Ziehl–Neelsen method, the Kinyoun technique does not require heating to facilitate penetration of the primary stain into the mycolic acid-rich cell envelope, reducing aerosol generation while providing reliable visualization of acid-fast organisms. It is commonly applied to cultured isolates and selected clinical specimens for microbiological diagnosis, biomedical research, histopathology, and educational applications.
