Static urine concentrators are passive ultrafiltration devices designed for clinical diagnostics. They enable centrifuge-free enrichment of low-abundance proteins from urine or cerebrospinal fluid (CSF) without the need for specialized equipment, making protein analysis more accessible and efficient.

Operating Principle

Absorbent pads positioned behind vertical ultrafiltration membranes create osmotic gradients that evaporate water over 30-120 minutes, achieving 50-200× concentration without centrifugation, vacuum, or heat. This makes the devices ideal for clinics lacking centrifuge rotors. PES membranes retain proteins larger than 7.5 kDa (e.g., Bence Jones proteins, light chains) while allowing salts and urea to pass, yielding analyzable concentrates directly via pipette.

Clinical Applications

The primary use of static urine concentrators is to enrich dilute urine samples for the diagnosis of multiple myeloma and AL amyloidosis through immunofixation electrophoresis (IFE), enabling detection of monoclonal light chains that may be missed in neat samples. CSF processing aids biomarker detection in conditions such as meningitis and multiple sclerosis, while bacterial antibody titer enhancement (e.g., H. influenzae, S. pneumoniae) supports latex agglutination assays. Compared to centrifugal methods, these devices offer reported savings of approximately 30% due to their simplicity and minimal equipment requirements.
