Histone extraction kits are laboratory reagent systems designed to isolate histone proteins from cells or tissues in a rapid and reproducible way. They are commonly used in epigenetics, chromatin biology, and nuclear protein analysis, especially when researchers need enriched histone fractions for downstream assays. Many kits are optimized to preserve histone post-translational modifications during extraction, which is important for studying acetylation, methylation, and other regulatory marks.

Principle of the Method

Most histone extraction kits use a sequence of cell lysis, nuclear enrichment, and acid-based or buffer-based histone solubilization steps. This approach separates histones from other cellular proteins while maintaining the integrity of the target molecules for analysis. Depending on the kit design, the protocol may be completed in about 1 hour, making it suitable for routine workflows and time-sensitive experiments.

Downstream Applications

Extracted histones are often used for Western blotting to examine global histone modification levels such as acetylation or methylation. They are also useful in enzyme-linked immunoassays and other post-translational modification assays that require purified histone input. For chromatin immunoprecipitation, however, a chromatin extraction kit is usually preferred rather than a histone purification kit.
