Ethyl acetate

Ethyl acetate is a widely used ester solvent in biochemistry, valued for its moderate polarity and rapid evaporation, making it ideal for extractions and chromatographic cleanups of biomolecules.

Chemical Properties

Ethyl acetate (CH₃COOCH₂CH₃) features an ester functional group with a planar carbonyl structure. It has a boiling point of 77.1°C, a melting point of -83°C, and a density of 0.902 g/mL. The solvent exhibits a characteristic fruity odor and is fully miscible with most organic solvents, while its solubility in water is limited (8.3 g/100 mL). Its dielectric constant (~6) positions it between polar protic and non-polar solvents. High volatility (vapor pressure 73 mmHg at 20°C) supports rapid, residue-free drying. Under basic conditions, ethyl acetate hydrolyzes into ethanol and acetic acid.

Biochemical Applications

In lipid biochemistry, ethyl acetate is commonly used to elute neutral lipids such as monoacylglycerols in thin-layer chromatography (TLC) following Folch extraction. It is also employed in the partitioning of flavonoids from plant metabolites using countercurrent distribution techniques.

In protein and peptide workflows, ethyl acetate can precipitate peptides from HPLC fractions as an alternative to acetone. In molecular biology, it facilitates the extraction of pigments or antibiotics from culture media without denaturing nucleic acids, preserving their integrity for downstream PCR applications. Additionally, it serves as a greener mobile phase in flash chromatography for the purification of carbohydrates and steroids.
