India ink stain

India ink stain is a classical negative staining technique extensively used in microbiology, mycology, and cell biology research to visualize encapsulated microorganisms without heat fixation or directly staining the microbial cells. Suspended carbon particles create a dark background while being excluded from the hydrated polysaccharide capsule, producing a characteristic clear halo surrounding encapsulated cells. In research laboratories, India ink staining remains an important, rapid, and inexpensive method for studying capsule morphology, capsule size variation, virulence-associated phenotypes, and host–pathogen interactions, particularly in Cryptococcus neoformans. It is frequently employed alongside culture, fluorescence microscopy, immunological assays, and molecular techniques to investigate fungal pathogenesis, capsule regulation, and environmental responses. Although modern antigen-based and molecular assays provide greater diagnostic sensitivity for clinical diagnosis, India ink continues to serve as a valuable research tool for direct microscopic examination, morphometric measurement of capsule size, and phenotypic characterization of encapsulated microorganisms. Because the technique relies on optical contrast rather than specific molecular labeling, it is primarily used to assess capsule morphology and size and does not provide information on capsule composition.
