| Description | Adrenomedullin (rat) is a 52-amino acid vasoactive peptide that functions as a potent endogenous regulator of vascular tone and cell proliferation in rat models. It binds with high affinity (Kd 0.3–0.6 nM) to specific receptors formed by calcitonin-receptor-like receptor and receptor-activity-modifying protein 2, activating cAMP signaling and p42/p44 ERK/MAP kinase pathways. In isolated perfused rat hearts, 1 nM and 10 nM adrenomedullin induces significant coronary vasodilation (-28% and -50%, respectively), while in cerebral microvessels it increases cAMP levels with more than 100-fold greater potency than CGRP. This compound is widely used in cardiovascular and neurovascular research to study receptor-mediated signaling, autocrine/paracrine growth regulation, and microcirculatory control mechanisms. |
| In vitro | Rat adrenomedullin (50-amino acid peptide) exhibits specific binding to rat tissues and cells. In Rat-2 fibroblasts, it binds with KD=0.43 nM and inhibits MAPK activity (PMID: 9931292). In rat lung membranes, binding IC50=5.8 nM; in heart, IC50=0.2 nM (PMID: 7720662). In pancreatic acini, it inhibits CCK-stimulated amylase secretion with IC50=1.1 pM (PMID: 9927317). In rat aorta, it induces endothelium-dependent relaxation with IC50=2.4 nM (PMID: 9605572). |
| In vivo | Adrenomedullin (rat) was identified as a key paracrine effector in adult VCAM1+ cardiac fibroblasts (aVCFs) that improved cardiac function post-MI in rat and mouse models. In vivo, aVCFs promoted cardiac lymphangiogenesis and cardioprotection; ADM knockdown attenuated these effects (PMID: 40957875). In cultured rat ventricular cardiac myocytes, hypoxia (1% O2) induced a time-dependent increase in ADM mRNA via HIF-1 consensus sites (PMID: 9651380). |

For example, if the intended dosage is 10 mg/kg for animals weighing 20 g , with a dosing volume of 100 μL per animal, and a total of 10 animals are to be administered, using a formulation of 
10% DMSO+ 40% PEG300+ 5% Tween 80+ 45% Saline/PBS/ddH2O , the resulting working solution concentration would be 2 mg/mL.Dissolve 2 mg of the compound in 100 μL DMSO to obtain a stock solution at a concentration of 20 mg/mL . If the required concentration exceeds the compound's known solubility, please contact us for technical support before proceeding.
1) Add 100 μL of the DMSO stock solution to 400 µL PEG300 and mix thoroughly until the solution becomes clear.
2) Add 50 µL Tween 80 and mix well until fully clarified.
3) Add 450 µL Saline,PBS or ddH2O and mix thoroughly until a homogeneous solution is obtained.
