Description
Lutein epoxide is a type of carotenoid that can be converted into lutein when exposed to light. It is utilized in research concerning plant photoprotection mechanisms.
In vitro
Method: Rat primary trigeminal neurons were co-incubated with RAMEB-Lutein epoxide (1, 3, 10, 100 μM) for 60 min. Intracellular Ca²⁺ influx induced by 200 μM mustard oil (MO) was measured using the Fura-2 ratiometric fluorescence method.
Result: Compared with the vehicle control group (29.03% ± 7.3%), the proportions of responsive neurons in the 3, 10, and 100 μM RAMEB-Lutein epoxide treatment groups decreased to 11.14% ± 2.85%, 3.1% ± 2.9%, and 4.8% ± 3.9%, respectively. The fluorescence increment (R) in the 100 μM treatment group was significantly reduced from 0.84 ± 0.21 to 0.21 ± 0.09, indicating a concentration-dependent inhibition of TRPA1 activation‑mediated calcium influx [1].
In vivo
Method: Mouse peritoneal macrophages were stimulated with LPS and co-treated with RAMEB-Lutein epoxide (1, 10, 100 μM) for 8 h. IL-1β levels in the supernatant were measured using ELISA.
Result: Compared with the LPS control group (853.2 pg/mL), 100 μM RAMEB-Lutein epoxide significantly inhibited LPS-induced IL-1β release. Inhibitory effects were also observed at 1 μM and 10 μM but did not reach statistical significance [1].
