Streptavidin-conjugated anti-horse primary antibodies are immunoreagents that combine specific antigen recognition with high-affinity biotin-binding capability for signal amplification and detection flexibility. Produced by immunizing hosts such as goat, rabbit, or mouse with purified horse immunoglobulins, the antibodies are affinity-purified and covalently conjugated to streptavidin, a 52 kDa tetrameric protein derived from Streptomyces avidinii. Each streptavidin molecule possesses four biotin-binding sites, each exhibiting exceptionally high affinity (Kd ≈ 10−14 M), among the strongest non-covalent interactions known in nature. The conjugation process typically couples 1–2 streptavidin molecules per antibody, preserving both antigen recognition and biotin-binding capacity. Unlike avidin, streptavidin lacks glycosylation, resulting in lower non-specific binding to biological membranes and reduced background in immunoassays. The streptavidin-antibody conjugate enables subsequent detection using any biotinylated reporter molecule, including biotinylated HRP, biotinylated alkaline phosphatase, biotinylated fluorophores (e.g., biotin-FITC, biotin-PE), or biotinylated gold nanoparticles, providing exceptional versatility. Extensive cross-adsorption against immunoglobulins from bovine, sheep, goat, mouse, and rabbit eliminates non-specific binding in multi-species samples.

Applications

Streptavidin-conjugated anti-horse antibodies are employed across diverse immunoassay formats where signal amplification is required. In immunohistochemistry and immunocytochemistry, these conjugates enable antigen detection in equine tissues and cultured cells using biotinylated chromogenic enzymes (HRP or alkaline phosphatase), providing intense staining with reduced background compared to direct enzyme conjugates. In ELISA, the streptavidin-antibody conjugate captures horse immunoglobulins and is subsequently detected with biotinylated HRP or biotinylated fluorophores, delivering amplified signal for quantifying low-abundance targets in serum, plasma, or cell culture supernatants. In Western blotting, these conjugates enable sensitive detection of equine proteins when combined with biotinylated HRP and chemiluminescent substrates, with typical amplification factors of 5- to 10-fold over direct conjugates. In flow cytometry and FACS, streptavidin conjugates detect biotinylated antigens or biotinylated secondary antibodies with enhanced sensitivity for immunophenotyping equine leukocytes and detecting surface markers. Additional applications include immunoprecipitation (using biotinylated capture antibodies), pull-down assays, and tissue microarrays. Working dilutions vary widely: 1:100–1:1,000 for immunohistochemistry, 1:1,000–1:20,000 for ELISA, and 1:1,000–1:10,000 for Western blotting.
