Introduction
This kit utilizes a DNA spin column and a novel solution system, suitable for the rapid and simple extraction of genomic DNA from plant samples containing polyphenols, polysaccharides, and enzyme inhibitors. It can purify DNA from one or more 100 mg fresh or 20 mg dried plant samples in 30 minutes. The extraction process does not require toxic organic solvents such as phenol or chloroform, nor time-consuming isopropanol or ethanol precipitation. It efficiently removes impurities such as polysaccharides, polyphenols, and enzyme inhibitors. The purified DNA is suitable for PCR, enzyme digestion, and hybridization experiments.
Fresh or dried plant tissue (cells) are ground and lysed; proteins, polysaccharides, and cell debris are precipitated and removed. Genomic DNA is then selectively adsorbed onto the silica membrane in the spin column under high salt conditions. A series of rapid wash-centrifugation steps further remove polysaccharides, polyphenols, cell metabolites, and proteins. Finally, a low-salt elution buffer elutes the purified genomic DNA from the silica membrane.

Assay Procedure Summary
- Grind plant tissue into a fine powder with liquid nitrogen.
- Transfer the powder to a tube with AP1 Buffer and RNase A.
- Vortex to homogenize and facilitate lysis.
- Add AP2 Buffer, mix, and centrifuge to remove impurities.
- Transfer the supernatant to a new tube and add AP3/E Buffer.
- Mix and transfer the mixture to a spin column.
- Centrifuge and discard the flow-through.
- Wash the spin column with Wash Buffer twice.
- Centrifuge to remove residual wash buffer.
- Elute the purified DNA with Elution Buffer.
