Allophycocyanin (APC) is a highly bright phycobiliprotein fluorophore, ideal for applications requiring high sensitivity and minimal background fluorescence. APC-conjugated anti-hamster antibodies are specifically designed to detect antigens or immunoglobulins of hamster origin. APC is efficiently excited by standard red lasers (633 nm or 640 nm) and emits fluorescence with a peak around 660 nm in the far-red spectrum. This spectral characteristic minimizes overlap with commonly used fluorophores such as FITC, PE, and Alexa Fluor® 488, making APC-conjugated antibodies excellent tools for multicolor flow cytometry panels and immunofluorescence applications.
Production, Purification, and Specificity
APC-conjugated anti-hamster antibodies are generally produced by immunizing a host species, most commonly goat, with purified hamster immunoglobulins. Following antibody generation, affinity chromatography is used to obtain highly purified antibodies, which are subsequently conjugated to APC under optimized conditions to achieve an appropriate fluorophore-to-protein ratio. To further improve specificity, many antibodies undergo cross-adsorption against serum proteins from other species, including mouse, rat, and human, thereby minimizing non-specific cross-reactivity. This process ensures reliable detection of hamster-derived targets in complex biological samples. APC-conjugated isotype control antibodies, such as Armenian hamster IgG controls, are also available to accurately assess background staining in flow cytometry experiments.
