1) Does the kit come with PBS -- is that what the included was solution is? Would Tris-HCL work as a wash solution? It is what my lab wrote they used in the pilot study but I do not have any context for why they chose it over PBS.
2) For getting tissue homogenates, do you happen to have any recommendations for grinding the tissue?
This kit (CSB-E08487f) is ready to use and provides all required reagents. No extra solution is required. Tris-HCl you mentioned may be not suitable for this kit. Please use the matched reagents of this kit, including the wash buffer.

Does this kit measure total cortisol levels, or only free/unbound cortisol? I will be using zebrafish tissue homogenates but I am not seeing any information in the protocol about whether it is total or only free cortisol.
This is a competitive kit, and it detects endogenous total cortisol. For zebrafish tissue samples, it is recommended to arrange a gradient pre-experiment.
