The prion protein is a large membrane protein that occurs normally in neurons of the human brain and is thought to be involved in synaptic transmission. In prion diseases, such as CJD, Gerstmann-Straussler-Scheinker syndrome (GSS), Fatal Familial Insomnia (FFI), Alpers Syndrome and Kuru, the normal cellular form of this protein (PrPc) is transformed into an altered protein when it comes into contact with an infectious prion protein (PrPsc) from another host.

Applications:
Suitable for use in ELISA and Western Blot. Other applications not tested.

Recommended Dilution:
ELISA: 1:1000-1:10,000
Western Blot: 1:500-1:2000
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Applications
Product Type: Mab|Isotype: IgG1,k|Clone No: 5G12|Host: mouse|Source: bovine|Concentration: As reported|Form: Supplied as a liquid in PBS/Casein-buffer, pH 8.0, 0.01% thimerosal. Immunogen
Peptide coresponding to aa225-228, Y236YQR239, from bovine bPrPc at C-terminal.
Form
Supplied as a liquid in PBS/Casein-buffer, pH 8.0, 0.01% thimerosal. Labeled with Biotin.
Purity
Purified by affinity chromatography.
Specificity
Recognizes bovine Prion Protein. Strong reactivity with recombinant PrPC from cattle, sheep and human, weak with mouse and hamster. Detects the native bovine PrPC and PrPSc in dot blots, the partly refolded recombinant bovine PrPC in ELISA, and denatured PrP in immunoblots of brain homogenates of healthy and BSE-affected animals.
