Bottle-top vacuum filters

Bottle-top vacuum filters are sterile, disposable filtration units designed for high-volume sterilization and clarification of tissue culture media, buffers, and biological solutions commonly used in biochemical and molecular biology laboratories. These filtration devices enable rapid and reliable removal of microorganisms and particulate contaminants while maintaining the sterility and integrity of sensitive biological preparations.

Filtration Mechanism

Under vacuum pressure, liquid samples pass through hydrophilic asymmetric membranes that provide high filtration flux and minimal hold-up volume. This mechanism allows efficient removal of bacteria, mycoplasma, and particulate matter while directing sterile filtrate into the receiving bottle. PES membranes generally provide the highest flow rates for aqueous solutions, PVDF membranes are suitable for filtration involving organic solvents, and SFCA membranes are frequently selected for sensitive cell culture applications due to their low extractable profiles.

Biochemical Applications

Bottle-top vacuum filters are widely used in cell culture and molecular biology workflows. In cell culture applications, 0.22 µm membranes are commonly employed to sterilize media such as FBS-supplemented DMEM or culture supernatants prior to downstream applications including cryopreservation or bioreactor feeding. In molecular biology and analytical workflows, 0.1 µm filters may be used for viral clarification or preparation of HPLC mobile phases, whereas 0.45 µm membranes are frequently used for clarification of cell lysates prior to protein purification. Their single-use design eliminates the need for cleaning validation and reduces contamination risks in regulated laboratory environments.
