Principle of the RSV ELISA Kit

The RSV ELISA (enzyme-linked immunosorbent assay) kit is based on the sandwich or indirect immunoassay principle. In a typical sandwich ELISA for RSV antigen detection, microplate wells are coated with monoclonal antibodies specific for RSV antigens. Patient samples containing RSV antigen bind to these antibodies. After washing, an enzyme-conjugated secondary antibody recognizing RSV antigen is added. A substrate solution is then applied that produces a colorimetric change proportional to the antigen concentration, measurable by optical density at 450 nm.

For antibody detection (IgG, IgM, or IgA), wells are coated with purified RSV antigens. Patient serum or plasma samples containing RSV-specific antibodies bind to antigens. After washing, enzyme-labeled anti-human immunoglobulin antibodies are introduced, followed by substrate development to quantify antibody levels.

The kits typically accept human serum, plasma, respiratory secretions, or other biological fluids. The assay is designed for in vitro diagnostic use and research applications, often supporting detection of acute infection (IgM) or immunity status (IgG).
