Qubit dsDNA HS (High Sensitivity) Assay Kit is a fluorescence-based quantitative assay designed specifically for accurate and selective measurement of double-stranded DNA (dsDNA) in biological samples. The assay is used in conjunction with Qubit Fluorometers, enabling DNA quantification in a wide concentration range from approximately 10 pg/μL to 100 ng/μL, making it highly suitable for sensitive applications requiring low DNA input detection.
Principle
The Qubit dsDNA HS assay utilizes a proprietary fluorescent dye that selectively binds to dsDNA over single-stranded DNA, RNA, proteins, free nucleotides, and other contaminants commonly found in samples. When the dye binds to dsDNA, it exhibits enhanced fluorescence intensity, which is measured by the fluorometer at an excitation wavelength near 485 nm and emission near 530 nm. The fluorescence signal is proportional to the dsDNA concentration, allowing precise quantification.
Performance and Applications
The assay demonstrates excellent selectivity for dsDNA, tolerating common contaminants such as salts, solvents, detergents, proteins, and RNA. Its high sensitivity allows detection of very low DNA quantities with coefficients of variation below 2%, ensuring reproducible results suitable for next-generation sequencing library quantification, PCR product quantification, and genomic or plasmid DNA analysis.
The Qubit dsDNA HS assay is widely utilized in molecular biology workflows requiring precise DNA input quantification to optimize downstream processes. Its ease of use, rapid turnaround, and reliability make it a gold standard for DNA quantification in research and clinical laboratories.
The Qubit dsDNA HS Assay Kit offers a robust, sensitive, and selective method for dsDNA quantitation. By providing a simple assay format combined with fluorometric measurement, it enables researchers to accurately quantify low abundance dsDNA samples, improving the quality and reproducibility of nucleic acid-based experiments.
