Glycerol

Glycerol is a fundamental polyol cryoprotectant and stabilizer widely used in biochemistry and molecular biology. It is valued for its high viscosity and strong hydrogen-bonding capacity, which help preserve biomolecular conformation and functional integrity under physical and chemical stress conditions.

Chemical Properties

Glycerol (C₃H₈O₃; 1,2,3-propanetriol) contains three hydroxyl groups attached to a three-carbon backbone. This structure confers complete miscibility with water, a boiling point of 290°C, and a dynamic viscosity of 1.49 Pa·s at 20°C. With a dielectric constant of approximately 42.5 and multiple hydrogen-bond donor and acceptor sites, glycerol interferes with ice crystal formation while maintaining hydration shells around macromolecules. It is non-toxic, hygroscopic, and laboratory-grade glycerol (>99% purity) ensures low endotoxin levels suitable for sensitive cell-based applications.

Biochemical Applications

In protein biochemistry, glycerol concentrations ranging from 10–50% are commonly used to stabilize enzymes during purification and long-term storage. It prevents protein aggregation at −20°C by promoting vitrification (glass-like state formation) and mitigates structural stress during freeze–thaw cycles or lyophilization.

In molecular biology, 5–15% glycerol is incorporated into PCR master mixes to enhance Taq polymerase processivity, particularly for GC-rich templates. It is also used at 5–10% in gel loading buffers to increase sample density and improve electrophoretic band resolution.

For lipid research, the glycerol backbone forms the structural basis of phospholipids in model membrane systems. In cryo-electron microscopy (cryo-EM), 2–10% glycerol is used to embed viruses and other macromolecular complexes while minimizing dehydration artifacts.
