Nerol

Nerol functions as the (Z)-cis isomer of geraniol, a key acyclic monoterpenoid alcohol involved in floral scent signaling, antimicrobial defense, and selective cytotoxicity. Its biological activity is primarily mediated through interactions with lipid membranes and modulation of reactive oxygen species (ROS), contributing to its role in plant physiology and potential biomedical applications.

Chemical Properties

Nerol ((Z)-3,7-dimethylocta-2,6-dien-1-ol, C₁₀H₁₈O, molecular weight 154.25 g/mol) is a colorless to pale yellow liquid with a boiling point of 225–227°C, density of 0.875–0.885 g/cm³, and refractive index (nD20) of 1.474. It is characterized by a sweet rose-neroli odor, distinct from geraniol due to its cis C2=C3 configuration. Nerol is lipophilic (logP 3.47) with limited water solubility (0.53 mg/mL). Chemically, it can undergo dehydration to form dipentene, isomerization to geraniol or linalool under acidic conditions, and oxidation to citral. Industrial synthesis is commonly achieved through β-pinene pyrolysis.

Biosynthetic Pathway

Nerol is biosynthesized via the plastidial methylerythritol phosphate (MEP) pathway, where geranyl diphosphate (GPP) is converted into neryl diphosphate and subsequently into nerol through the action of neryl diphosphate synthase (NDPS) and alcohol dehydrogenases, or by direct hydrolysis. It is naturally present in essential oils such as neroli oil (0.5–2%), rose otto (5–15%), and lavender. Emission rates in plants during anthesis typically range from 1 to 10 μg·g⁻¹·h⁻¹ (dry weight). Advances in metabolic engineering have enabled microbial production, with recombinant Escherichia coli strains expressing NDPS and alcohol dehydrogenases achieving fermentation titers of 100–500 mg/L.
