Descripciòn

Fluorescent DOTAP (NBD-DOTAP) is a cationic lipid which can be used for nucleic acid and protein delivery. Fluorescent DOTAP is labeled with a fluorophore NBD (maximum excitation/emission wavelength ∼463/536 nm). Fluorescent DOTAP can be used for gene delivery systems, drug delivery, as well as cell imaging and nanocarrier tracking. Fluorescent DOTAP is an ideal candidate for both biological and pharmaceutical formulation (e.g. co-delivery of vaccines) research[1][3].

In Vitro

Fluorescent DOTAP results in a higher hemolysis with increasing concentration of DOTAP in fusogenic liposomes[2].

In Vivo

Fluorescent DOTAP (0.5% (w/w), s.c., one single dose) increases the numbers of CD11c+ cells among total lymph node cells more than 2.5-fold compared to the untreated at 4 h in C57BL/6 female mice[3].

Animal Model: C57BL/6 female mice (6-7 weeks old) with subcutaneous tumors[3]
Dosage: 0.5% (w/w)
Administration: Subcutaneous injection (s.c.), one single dose
Result: Showed neither CD8+ or CD8- cells took up NBD after fluorescent DOTAP injection.
Demonstrated the main uptake is by DC (80%) soon after immunization.
Induced migration of activated DC to the DLN.
Resulted in DC-T cell interactions and eliciting T cell responses.

For the following dissolution methods, please ensure to first prepare a clear stock solution using an In Vitro approach and then sequentially add co-solvents:
To ensure reliable experimental results, the clarified stock solution can be appropriately stored based on storage conditions. As for the working solution for in vivo experiments, it is recommended to prepare freshly and use it on the same day.
The percentages shown for the solvents indicate their volumetric ratio in the final prepared solution. If precipitation or phase separation occurs during preparation, heat and/or sonication can be used to aid dissolution.

  • Protocol 1

    Add each solvent one by one:  10% DMSO    90% (20% SBE-β-CD in Saline)

    Solubility: ≥ 2 mg/mL (2.82 mM); Clear solution

    This protocol yields a clear solution of ≥ 2 mg/mL (saturation unknown).

    Taking 1 mL working solution as an example, add 100 μL DMSO stock solution (20.0 mg/mL) to 900 μL 20% SBE-β-CD in Saline, and mix evenly.

    Preparation of 20% SBE-β-CD in Saline (4°C, storage for one week): 2 g SBE-β-CD powder is dissolved in 10 mL Saline, completely dissolve until clear.
